I had an interview a couple of days ago and I am following up with a hand written letter. I followed MT recommendations,I felt the interview went well. However, there was one question I did not fully nail. I am planning to address this in my letter; my response will be short and to the point. I’m looking for some advice as to whether this is a good idea or not.

Hi,
I interviewed a candidate a few weeks ago. He had a great interview, except for one answer... And I forgot all about the one bad answer! I was ready to make an offer, when he sent an email about his one poor answer, which I had forgotten. That got me thinking again.
If you make a mistake, so what. Everybody does, nobody is perfect. Unless it's a critical flaw, I would not recommend bringing it up again.

 I'd agree with Finnallred (above).  
The purpose of the follow up letter is to say thank you, to reinforce your interest and to add to the positive, professional impression you made at interview. I don't see it as a continuation of the interview and, in my opinion,  busy recruiters probably wouldn't want it to be.
 
You might be agonising over this now but the recruiter probably isn't, so I'd be more concerned about getting the thank you off promptly and not trying to do more than that.

In the end I decided not to bring up the question. Judging by the commnets that was the right call. Glad to see we're on the same page.
